{"id":5699,"date":"2018-10-19T09:30:09","date_gmt":"2018-10-19T14:30:09","guid":{"rendered":"https:\/\/cardiologynownews.org\/?p=5699"},"modified":"2018-10-19T09:30:42","modified_gmt":"2018-10-19T14:30:42","slug":"assessment-of-genomic-metagrs-scoring-system-in-prediction-of-coronary-artery-disease","status":"publish","type":"post","link":"https:\/\/cardiologynownews.org\/?p=5699","title":{"rendered":"Assessment of Genomic MetaGRS Scoring System in Prediction of Coronary Artery Disease"},"content":{"rendered":"<p>A study published by <em><a href=\"http:\/\/www.onlinejacc.org\/content\/72\/16\/1883\">Journal of the American College of Cardiology<\/a><\/em> described a genomic score having the potential to stratify the risk of coronary artery disease (CAD) and emphasized the possible use of\u00a0genomic\u00a0screening in early life, adding to traditional risk factors.<!--more--><\/p>\n<p>Inoyue et al used UK biobank to collect comprehensive data on &gt;480,000 individuals. UK Biobank is an open access, large, multicenter, population-based prospective study with the purpose of allowing investigation of genetic and non-genetic determinants. Patient populations consist of individuals aged between 40 to 69 years at recruitment across the United Kingdom between 2006 and 2010. Information was gathered using a standardized questionnaire on sociodemographic characteristics, health status and physician-diagnosed medical conditions, family history, and lifestyle factors. They used a meta-analysis strategy to develop a genomic risk score for CAD.<\/p>\n<p><img loading=\"lazy\" loading=\"lazy\" decoding=\"async\" class=\" wp-image-5730 alignleft\" src=\"https:\/\/cardiologynownews.org\/wp-content\/uploads\/2018\/10\/InouyeM.jpg\" alt=\"\" width=\"304\" height=\"304\" srcset=\"https:\/\/cardiologynownews.org\/wp-content\/uploads\/2018\/10\/InouyeM.jpg 330w, https:\/\/cardiologynownews.org\/wp-content\/uploads\/2018\/10\/InouyeM-300x300.jpg 300w\" sizes=\"auto, (max-width: 304px) 100vw, 304px\" \/>[perfectpullquote align=&#8221;full&#8221; bordertop=&#8221;false&#8221; cite=&#8221;&#8221; link=&#8221;&#8221; color=&#8221;&#8221; class=&#8221;&#8221; size=&#8221;&#8221;]<strong>&#8220;The genomic score developed and evaluated in the\u00a0present study strengthens the concept of using\u00a0genomic information to stratify individuals for CAD\u00a0risk in general populations and demonstrates the\u00a0potential for genomic screening in early life to complement\u00a0conventional risk prediction.\u00a0Although applied health studies will be needed to\u00a0evaluate properly the clinical utility of CAD genomic\u00a0risk scores, elements of potential clinical implementation\u00a0can now be foreseen.&#8221;-Dr.\u00a0Michael Inouye, Ph.D.<\/strong>[\/perfectpullquote]<br \/>\nA total of 9,729 cases of CAD were present at the time of recruitments and 12,513 new cases were diagnosed during a mean follow-up period of 6.2 years. The meta-analysis performed by Inoyue et al resulted in genomic risk score \u201cmetaGRS\u201d which included 1,745,180 genetic variants and explained that 26.8% of CAD might be contributed by heredity. Also, they suggested that metaGRS had a fundamentally stronger association with CAD risk according to the hazard ratio (95% confidence interval [CI]: 1.68 to 1.73) and positive predictive value (PPV) when compared to other external validation studies.<\/p>\n<p>The authors also acknowledged that the relationship of the metaGRS with traditional clinical risk scores was not assessed as lipid measurements were not available in the UK Biobank.
